Objective:To observe the effect of tuina pressing and kneading method on motor function and the expression of NF-κB and c-Fos in rats with sciatic nerve injury.Methods:A total of thirty-six SPF rats were randomly divided into blank group,model group and tuina group,with 12 rats in each group.A rat model of chronic constriction injury was established in the model and tuina groups,while the blank group did not receive any intervention.On the fourth day after successful modeling,the tuina group rhythmi-cally pressed and kneaded the area around the"Weizhong acupoint"for 10 minutes each time,once a day for 14 days,while the blank and model groups did not receive any intervention.The TreadScan gait analysis system was used to detect sciatic nerve function index and gait parameters in rats before modeling and at 7 and 14 days of intervention;HE staining method was used to observe pathologi-cal changes in the sciatic nerve;Western blot was used to detect the protein expression of NF-κB and c-Fos in the sciatic nerve.Re-sults:The HE staining results showed that the sciatic nerve fibers in the model group had poor continuity,disordered arrangement,en-larged fiber gaps,partial demyelination,axonal swelling,degeneration,and even rupture,and increased distribution of inflammatory cells;the arrangement of sciatic nerve fibers in the tuina group was neat and tight compared to the model group,with some myelin sheaths and axons regenerating and a decrease in inflammatory cells.Compared with the blank group,the model group showed a sig-nificant decrease in sciatic nerve function index,affected limb stride,average walking speed,and standing pressure on the 7th and 14th day of intervention(P<0.05),a significant reduction in standing time(P<0.05),and a significant increase in sciatic nerve NF-κB and c-Fos protein expression(P<0.05).Compared with the model group,the tuina group showed a significant increase in sciatic nerve function index,affected limb stride,average walking speed,and standing pressure on the 7th and 14th day of intervention(P<0.05),a significant prolongation of standing time(P<0.05),and a significant decrease in sciatic nerve NF-κB and c-Fos protein expression(P<0.05).Conclusion:Tuina pressing and kneading method may reduce local inflammation level after sciatic nerve injury by downregu-lating the protein expression of NF-κB and c-Fos,and promote the repair of the injured nerve,so as to improve the motor function of the affected lower limb.
