Objective:To investigate the effect of Babaodan(BBD)on xenografted breast tumor in mice based on nuclear factor-κB(NF-κB)signaling pathway.Methods:A total of twelve 6-week-old female BALB/c nude mice with SPF grade were subcutane-ously injected with breast cancer cells MDA-MB-231 to construct a xenografted breast tumor mouse model and were randomly divid-ed into control group and intervention group,with 6 mice in each group.The intervention group was given BBD by gastric lavage at a dose of 5 g/(kg·d),while the control group was given physiological saline by gavage at a dose of 0.1 mL/(kg·d)for 28 consecutive days.The body weight and tumor volume of mice were measured every 2 days after intervention,and the tumor weight was measured before and after intervention.HE staining method was used to observe pathological changes in heart,liver,and kidney tissues;ELISA was used to detect the levels of serum creatine kinase(CK),lactate dehydrogenase(LDH),aspartate aminotransferase(AST),alanine aminotransferase(ALT),tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6);immunohistochemistry was used to detect the pro-tein expression of antigenic nuclear proliferation factor Ki67,proliferating cell nuclear antigen(PCNA),NF-κB p65 subunit(p65),p-p65,NF-κB inhibitory protein-α(IκB-α),and p-IκB-α in breast tumor tissues.Results:The HE staining results showed that the mor-phology of the heart,liver,and kidney tissues in both groups was normal,with normal cell morphology.No inflammatory cell infiltra-tion,cell necrosis,or other toxicity related pathological changes were observed.There was no significant difference in body weight be-tween the two groups of mice at different time periods(P>0.05).There was no significant difference in tumor volume between the two groups within 14 days(P>0.05),and the tumor volume of the intervention group decreased significantly compared to the control group from day 16 onwards(P<0.05).Compared with the control group,the tumor weight and serum LDH content in the intervention group significantly reduced(P<0.05),and the positive expression rates of Ki67 and PCNA and the ratios of p-p65/p65 and p-IκB-α/IκB-α in tumor tissues significantly decreased(P<0.05).There was no significant difference in the contents of CK,AST and ALT be-tween the two groups(P>0.05).Conclusion:BBD can inhibit the growth and inflammatory reaction of xenografted breast tumor in mice,and its mechanism may be related to the regulation of NF-κB signaling pathway.
