丹参木质素及其单体含量的测定

本文建立了一种准确、快速的检测丹参中木质素及其单体含量的方法.采用Klason法和紫外分光光度法分别对丹参根和茎中酸不溶性木质素(Klason木质素)和酸溶性木质素含量进行了测定;运用硫代酸解法并结合气相色谱-质谱法(GC-MS)分别对丹参根和茎中各木质素单体组成进行了分析.结果显示,总木质素在丹参根和茎中的含量分别为280.7mg/g和160.4mg/g,其中Klason木质素含量分别为173.8mg/g和134.5 mg/g;酸溶性木质素含量分别为106.9mg/g和25.9 mg/g.H型、G型和S型木质素单体在丹参根中的含量分别为14.4μmol/g、2 070.2 μmol/g、1 886.4μmol/g,在丹参茎中的含量分别为15.3 μmol/g、212.8 μmol/g、50.6 μmol/g.利用本研究建立的方法可以准确、快速的检测丹参中各类木质素及其单体的含量,此方法可为其它药用植物木质素类化合物的定性、定量分析提供借鉴.
Determination of Lignin Content and Lignin Monomer Composition in Salvia miltiorrhiza Bge
An accurate, rapid method for determining lignin content and lignin monomer composition in Salvia miltiorrhiza Bge. was established. Acid-insoluble lignin(Klason lignin) and acid-soluble lignin in roots and stems of S. miltiorrhiza were measured by Klason method and UV spectrophotometry, respectively. The lignin monomer in roots and stems of S. miltiorrhiza was extracted by a streamlined thioacidolysis procedure and analyzed via gas chromatography-mass spectrometry(GC-MS), Total lignin content in roots and stems of S. miltiorrhiza were 280. 7 mg/g and 160, 4 mg/g,in which the Klason lignin content was 173. 8 mg/g and 134. 5 mg/g and acid-soluble lignin content was 106. 9 mg/g and 25, 9 mg/g,respectively. The main p-hydroxyphenyl ( H), guaiacyl (G), and syringyl(S) lignin monomer content were 14. 4 μmol/g.2 070, 2 /μmol/g and 1 886. 4 μmol/g in roots,and 15. 3 μmol/g,212. 8 μmol/ g and 50. 6 μmol/g in stems,respectively. The results showed that lignin and its monomer lignin content could be accurately and quickly determined by the method used in this study,and this method might provide reference for the qualitative and quantitative analysis of lignin composition in other medicinal plants.
