Fe3O4@ZIF-8-表面辅助激光解吸/电离质谱法检测美洲大蠊多肽

本实验在室温下合成了以Zn作为金属中心,2-甲基咪唑作为连接剂的磁性金属有机框架材料Fe3O4@ZIF-8,以Fe3O4@ZIF-8作为固体基质,通过建立Fe3O4@ZIF-8-表面辅助激光解吸/电离质谱(SALDI-MS)法,实现了美洲大蠊小分子多肽IPMTAPGL-NH2、LTAPGL-NH2、SLMTGPGL-NH2、SLHTAPGL-NH2 的快速检测.实验优化了基质浓度、检测限和点样方法等条件,考察了该基质对美洲大蠊多肽IPMTAPGL-NH2的质谱信号影响.与传统基质辅助激光解析电离质谱法(MALDI-MS)相比,Fe3O4@ZIF-8-SALDI-MS法检测背景更干净,目标多肽响应更强,检测灵敏度更高.该法可应用于血样中美洲大蠊多肽IPMTAPGL-NH2的快速检测.
Detection of Periplaneta Americana Peptide IPMTAPGL-NH2 by Fe3O4@ZIF-8-surface-assisted Laser Desorption/ionization Mass Spectrometry
Peptides from Periplaneta americana are not thermally stable,and the conventional detection methods are often difficult to achieve the desired state.In this experiment,magnetic metal-organic frameworks(MMOFs)Fe3O4@ZIF-8 were synthesized at room temperature,and Fe3O4@ZIF-8 was used as a solid matrix to establish the Fe3O4@ZIF-8-surface-assisted laser desorption/ionization mass spectrometry(SALDI-MS)method,which was applied for the rapid detection of the small molecule peptides from Periplaneta americana,such as IPMTAPGL-NH2,LTAPGL-NH2,SLMTGPGL-NH2 and SLHTAPGL-NH2.The matrix effect on mass spectrometric signal of the peptides from Periplaneta americana IPMTAPGL-NH2 was investigated by optimizing the matrix concentration,detection limit and spotting method.Compared with matrix-assisted laser desorption/ionization-mass spectrometry(MALDI-MS),Fe3O4@ZIF-8-SALDI-MS method has a cleaner background,stronger response and higher detection sensitivity.This method can be applied to the rapid detection of peptide IPMTAPGL-NH2 from Periplaneta americana in blood samples.
