Detection of Periplaneta Americana Peptide IPMTAPGL-NH2 by Fe3O4@ZIF-8-surface-assisted Laser Desorption/ionization Mass Spectrometry
Peptides from Periplaneta americana are not thermally stable,and the conventional detection methods are often difficult to achieve the desired state.In this experiment,magnetic metal-organic frameworks(MMOFs)Fe3O4@ZIF-8 were synthesized at room temperature,and Fe3O4@ZIF-8 was used as a solid matrix to establish the Fe3O4@ZIF-8-surface-assisted laser desorption/ionization mass spectrometry(SALDI-MS)method,which was applied for the rapid detection of the small molecule peptides from Periplaneta americana,such as IPMTAPGL-NH2,LTAPGL-NH2,SLMTGPGL-NH2 and SLHTAPGL-NH2.The matrix effect on mass spectrometric signal of the peptides from Periplaneta americana IPMTAPGL-NH2 was investigated by optimizing the matrix concentration,detection limit and spotting method.Compared with matrix-assisted laser desorption/ionization-mass spectrometry(MALDI-MS),Fe3O4@ZIF-8-SALDI-MS method has a cleaner background,stronger response and higher detection sensitivity.This method can be applied to the rapid detection of peptide IPMTAPGL-NH2 from Periplaneta americana in blood samples.
