Construction of Fluorescent Silver Nanoclusters Using Captopril as Template for Detecting the Biothiols
Biothiols play an important role in the metabolism of organisms,such as the abnormal content of cysteine(Cys)and glutathione(GSH)in human body will lead to a series of diseases.To establish a rapid and quantitative fluorescence assay method for detecting biothiols,red fluorescent silver nanoclusters was synthesized by wet chemical reduction method using captopril(Capt)as a template(namely Capt-AgNCs).The morphology,structure,elemental composition and optical properties of Capt-AgNCs were characterized.A new method for quantitative detection of Cys and GSH was established based on the fluorescence quenching effect of biothiols on Capt-AgNCs.Under the optimal experimental conditions,the linear ranges for Cys and GSH were 1-100 μmol/L and 2-180 μmol/L,respectively.And the detection limits for Cys and GSH were 0.054 and 0.23 μmol/L,respectively.The method could be used for the determination of Cys in human plasma with the recoveries of 95.3%-103.3%.The experimental results showed that Capt-AgNCs as a novel signal-off fluorescent probe could be used for detection of biothiols such as Cys and GSH.
