Simultaneous Determination of 6 Nucleosides in Arabidopsis thaliana by High Performance Liquid Chromatography
A high performance liquid chromatography(HPLC)method was established for the simultaneous determination of 6 nucleosides(thymidine,2'-deoxyadenosine,adenosine,2'-deoxyguanosine,cytidine,guanosine)in Arabidopsis thaliana.After pretreatment of Arabidopsis thaliana,the target compound was separated using a Venusil XBP C18 column(250 mm×4.6 mm×5 μm)by gradient elution with water-methanol as the mobile phase.The flow rate was 0.5 mL/min,the column temperature was 35 ℃,and the analysis was performed at 260 nm wavelength.The results showed that the 6 nucleosides had a good linear relationship(R2 ≥0.999)within the range of 0.01-1 000 μg/mL,the detection limit was 0.10-0.16 mg/kg,and the spiked recovery was 70.70%-103.09%.The method has the advantages of simple operation,good precision,strong repeatability and high accuracy.It can simultaneously determine the content of 6 nucleosides in Arabidopsis thaliana.
