Determination of Four Natural Carotenoids in Tomato Flesh by High Performance Liquid Chromatography
A new method for the quantitative determination of four lipophilic natural carotenoids in tomato pulp by high performance liquid chromatography(HPLC)was established.This method used YMC C30 chromatographic column as the stationary phase,and methanol,acetonitrile and isopropanol as the mobile phase.The methodology has been validated by the determination of lutein,zeaxanthin,α-carotene and β-carotene in tomato pulp.Under this chromatographic condition,the four natural carotenoids had a high linear correlation,the correlation coefficient(R2)was higher than 0.9980,the detection limit(LOD)of the four colorants was between 0.05 and 0.1 μg/mL,the limit of quantification(LOQ)was in the range of 0.1-0.3 μg/mL,and the recovery rate of the four pigments was all within 85.50%-112.4%.The method has the advantages of high flow sensitivity,good repeatability,high recovery rate and high instrument penetration rate.It is suitable for the analysis of natural carotenoids in fruits and vegetables.
