高效液相色谱法测定番茄果肉中4种天然类胡萝卜素含量

建立了一种高效液相色谱法测定番茄果肉中4种亲脂性天然类胡萝卜素的方法.该方法选择YMC C30色谱柱,以甲醇、乙腈和异丙醇为流动相,对番茄果肉中叶黄素、玉米黄素、α-胡萝卜素和β-胡萝卜素4种色素的含量测定进行了方法验证.在优化的色谱条件下,4种亲脂性天然类胡萝卜素线性相关性良好,相关系数(R2)均高于0.9980;检出限(LOD)在0.05~0.10 μg/mL之间,定量限(LOQ)在0.10~0.30 μg/mL之间;回收率在85.5%~112.4%之间.该方法操作简单、准确、可靠,适合蔬菜水果中天然类胡萝卜素含量分析.
Determination of Four Natural Carotenoids in Tomato Flesh by High Performance Liquid Chromatography
A new method for the quantitative determination of four lipophilic natural carotenoids in tomato pulp by high performance liquid chromatography(HPLC)was established.This method used YMC C30 chromatographic column as the stationary phase,and methanol,acetonitrile and isopropanol as the mobile phase.The methodology has been validated by the determination of lutein,zeaxanthin,α-carotene and β-carotene in tomato pulp.Under this chromatographic condition,the four natural carotenoids had a high linear correlation,the correlation coefficient(R2)was higher than 0.9980,the detection limit(LOD)of the four colorants was between 0.05 and 0.1 μg/mL,the limit of quantification(LOQ)was in the range of 0.1-0.3 μg/mL,and the recovery rate of the four pigments was all within 85.50%-112.4%.The method has the advantages of high flow sensitivity,good repeatability,high recovery rate and high instrument penetration rate.It is suitable for the analysis of natural carotenoids in fruits and vegetables.
