Determination of Periplaneta Americana Peptide by Electrochemical Immunosensor Based on Ag-TiO2-CS Nanocomposites
In this study,a new method for the electrochemical detection of Periplaneta Americata peptide(IPMTAPGI-NH2)was developed.In the experiment,nano-silver-titanium dioxide-chitosan(Ag-TiO2-CS)composite was used as signal amplification material to modify the electrode.Further,the antibody was fixed on the gold electrode modified with Ag-TiO2-CS nanocomposite by chitosan embedding method,and a simple labeling free electrochemical immune sensor was prepared by the specific reaction between the peptide and the antibody.Under the optimal experimental conditions,the linear range of the designed electrochemical immune sensor ranged from 1 to 25 ng/mL,and the detection limit was 0.83 ng/mL.The established method was applied for the detection of the peptide in the Kangfuxin solution,a preparation of Periplaneta Americana.The recovery was between 94.97%and 108.1%.The electrochemical immunosensor constructed in this study is simple,rapid and sensitive,and can be used for the determination of the polypeptide in the preparation of Periplaneta Americana.
