基于轨道阱高分辨质谱技术建立水产品中硝基呋喃类代谢物的快速筛查方法

本文采用超高效液相色谱-四极杆/轨道阱高分辨质谱法,建立了水产品中4种硝基呋喃类代谢物的快速筛查方法,同时建立质谱数据库,摆脱标准品依赖,实现高通量筛查.硝基呋喃类代谢物在50 ℃下进行衍生化反应,以Hypersil GOLD色谱柱(100 mm×2.1 mm,1.9 μm)实现分离,采用全扫描自动触发二级扫描模式(Full MS/dd MS2)对呋喃唑酮、呋喃它酮、呋喃妥因和呋喃西林进行测定.方法的线性范围为1~100 ng/mL,相关系数大于0.99,检出限为0.5 μg/kg,回收率在62.5%~113.3%,相对标准偏差在2.6%~13.5%.所建立方法前处理速度快、灵敏度高、重现性好,可用于水产品中硝基呋喃类代谢物的快速筛查.
A Rapid Screening Method Based on High Resolution Orbit-Trap Mass Spectrometry for Detection of Fornitrofuran Metabolites in Aquatic Products
A rapid screening method for detection of four kinds of nitrofuran metabolites in aquatic products was established by ultra high performance liquid chromatography-quadrupole/orbital trap high resolution mass spectrometry,and a mass spectrum database was established to realize high throughput screening without standard substances.Nitrofuran metabolites were derivatized at 50℃ and separated by Hypersil GOLD column.Furazolidone,furfuratone,furantoin and furacillin were detected by Full MS/dd MS2.The linear range was 1-100 ng/mL,the correlation coefficients were greater than 0.99,the detection limits were 0.5 μg/kg,the recoveries were 62.5%-113.3%,and the relative standard deviations were 2.6%-13.5%.The pretreatment is rapid,sensitive and reproducible,and could be used for the rapid screening of poppy husk alkaloids in food.
