The relationship between serum Atg5,Atg12,LC3 levels and the severity of disease and prognosis in patients with sudden deafness
Objective To investigate the relationship between the levels of serum autophagy re-lated gene 5(Atg5),autophagy related gene 12(Atg12),microtubule-associated protein Ⅰ light chain 3(LC3)and the severity and prognosis of sudden deafness(SD)patients.Methods 200 SD patients admitted to Xing-tan Hospital Affiliated with the Shunde Hospital of Southern Medical University from January 2021 to Decem-ber 2023 were selected as the SD group.Additionally,80 volunteers who underwent physical examinations dur-ing the same period were selected as the control group.Serum levels of Atg5,Atg12,and LC3 were tested in both groups.The SD patients were further categorized based on their pure tone audiometry(PTA)results into four subgroups:mild hearing loss,moderate hearing loss,severe hearing loss and extremely severe hearing loss.They were also divided into two groups based on their prognosis:poor prognosis and good prognosis.Lo-gistic regression was used to analyze the prognostic factors of SD patients.Results The serum levels of Atg5,Atg12,and LC3 in the SD group were lower than those in the control group,and the difference was statisti-cally significant(P<0.05).The serum levels of Atg5,Atg12,and LC3 in the SD group were found to be the following orders:mild hearing loss subgroup>moderate hearing loss subgroup>severe hearing loss subgroup>extremely severe hearing loss subgroup,and the difference was statistically significant(P<0.05).Addition-ally,the serum levels of Atg5,Atg12,and LC3 in the SD group were negatively correlated with PTA(P<0.05).Within the SD group,patients with a poor prognosis had higher ages,longer disease courses,and a higher proportion of total deafness compared to those with a good prognosis.Furthermore,the serum levels of Atg5,Atg12,and LC3 were lower in patients with poor prognosis compared to those with a good prognosis,with statistical significance(P<0.05).Logistic regression analysis revealed that increased age,prolongation of disease course,total deafness and decreased serum Atg5,Atg12,and LC3 levels were independent influencing factors for the poor prognosis of SD patients(P<0.05).Conclusion The levels of serum Atg5,Atg12,and LC3 in SD patients were found to be decreased and correlated with the severity of the disease and prognosis.
