摘要
目的 探讨流式细胞术在肿瘤细胞学中的检测效果及特征.方法 选自本院于2015年3月至2017年3月期间收治的肿瘤患者194例作为观察组,另选自本院于2015年3月至2017年3月期间健康体检者147例作为对照组.分别取两组外周静脉血,制成单细胞悬液,应用BD FACSCanto流式细胞仪计算淋巴细胞中各标记细胞的百分率.比较两组外周血T淋巴细胞亚群抗原变化、B淋巴细胞抗原变化及NK细胞和CD3+/CD25+变化.结果 观察组CD3+(56.82±4.15)%、CD4+ (31.20±3.17)%、CD4+/CD8+(0.95±0.18)低于对照组CD3+(67.29±5.67)%、CD4+(35.68±4.36)%、CD4+/CD8+(1.40±0.29),而CD8+(32.97±3.76)%高于对照组(25.41±2.30)%,差异均有统计学意义(均P<0.05).观察组CD19+(7.95±1.74)%和CD20+(8.41±1.90)%低于对照组CD19+(12.84±2.57)%和CD20+(13.09±2.93)%,差异有统计学意义(P<0.05).观察组NK细胞百分比(26.42±3.15)%高于对照组(18.65±2.24)%,差异有统计学意义(P<0.05);而两组CD3+/CD25+比较无统计学差异(P>0.05).结论 流式细胞术检测肿瘤患者存在细胞免疫异常,特征主要为外周T淋巴细胞亚群CD3+、CD4+、CD4+/CD8+升高而CD8+降低,CD19+和CD20+降低,及NK细胞降低,具有重要的研究价值.
Abstract
Objective:To investigate the effect of flow cytometry in detection of tumor cytology and its characteristics.Methods 194 patients with cancer treated at our hospital from March,2015 to March,2017 were selected as an observation group and 147 cases taking physical examination at our hospital from March,2015 to March,2017 a control group.Peripheral venous blood were collected from both groups and made into single cell suspension.The percentage of labeled cells in lymphocytes was calculated by BD FACSCanto.The changes of peripheral blood T lymphocyte subsets,antigen,B lymphocyte antigen,NK cell,and CD3+/CD25+were compared between the two groups.Results The CD3+,CD4+,CD4+/CD8+,CD19+,and CD20+ were lower and the CD8+ and the percentage of NK cells were higher in the observation group than in the control group [(56.82±4.15) % vs.(67.29±5.67) %,(31.20±3.17) % vs.(35.68±4.36) %,(0.95±0.18) vs.(1.40±0.29),(32.97±3.76) % vs.(25.41+2.30) %,(7.95±1.74) % vs.(12.84±2.57) %,(8.41±1.90) % vs.(13.09±2.93) %,and (26.42±3.15) % vs.(18.65±2.24) %],with statistical differences (all P<0.05).There was no statistical difference in CD3+/CD25+ between the two groups (P>0.05).Conclusions Flow cytometry can detect the abnormal cellular immunity of tumor patients.The main features are that CD3+,CD4+,CD4+/CD8+,CD19+,and CD20+ decrease and CD8+ and NK cells increase.Therefore,it has important research value.
