组蛋白甲基转移酶2在常见肿瘤中的机制及临床研究进展

摘要
组蛋白甲基转移酶2(SET and MYND domain-containing proteins 2,SMYD2)是SMYD家族成员,其功能是对组蛋白和非组蛋白进行甲基化修饰,调节下游的靶基因和信号通路,广泛参与肿瘤、炎症及免疫失调等疾病.近年来,恶性肿瘤的发病率呈逐年增高的趋势,严重影响患者的生活质量,给家庭和社会带来极大的经济负担.肿瘤发生的潜在机制在很大程度上仍不清楚,但越来越多的证据表明甲基化的异常调节与肿瘤发生密切相关.SMYD2参与多个信号通路的调控进而影响肿瘤干细胞、肿瘤细胞增殖与转移以及耐药等生物学特征.因此,本文对SMYD2在常见肿瘤中的作用机制进行综述,以期为SMYD2的机制研究及其抑制剂用于肿瘤治疗提供理论基础.
Abstract
SET and MYND domain-containing proteins 2(SMYD2)is an important histone methyltransferase belonging to the SMYD family.Its function is to methylate and modify histone and non-histone proteins,regulate downstream target genes and signal pathways,and widely participate in tumors,inflammation,immune disorders,and other diseases.In recent years,the incidence rate of malignant tumors has been increasing year by year,affecting patients'quality of life and imposing a significant financial burden on families and society.The underlying mechanism of tumorigenesis is still unknown,but increasing evidences suggest that abnormal methylation regulation is linked to tumorigenesis.SMYD2 regulates multiple signal pathways,which affects the biological properties of tumor cell proliferation and metastasis,cancer stem cells,and drug resistance.This article reviews the mechanism of SMYD2 in common tumors in order to provide a theoretical basis for SMYD2 and its inhibitor for tumor treatment.
