摘要
桥粒(Desmosome,DSM)是大型的大分子蛋白质集合体,它将中间丝细胞骨架与钙黏蛋白介导的细胞黏附点机械地连接起来,为维持经历机械应力组织(如皮肤、心肌和胃肠道黏膜等)提供结构完整性.桥粒胶蛋白(Desmocollins,DSCs)属于钙黏蛋白超基因家族,在细胞间黏附中发挥重要作用.桥粒胶蛋白2(Desmocollin-2,DSC2)是DSC家族成员,已被发现在几种类型的癌症中异常表达,影响肿瘤细胞的侵袭和转移,并参与肿瘤进展.DSC2在人类不同疾病中的表达和调控机制的差异为其治疗提供了潜在靶点.研究并阐明DSC2在肿瘤中的发生机制,探索临床检查和治疗的具体靶点,可为疾病的早期治疗提供实用思路.本文对DSC2的分子结构及其相关分子机制、DSC2与肿瘤的关系进行综述.
Abstract
Desmosomes(DSM)are large assemblies of macromolecules that mechanically link the cytoskeleton of intermediate filaments to cadherin-mediated cell adhesion sites,providing structural integrity for the maintenance of mechanically stressed tissues such as skin,myocardium and gastrointestinal mucosa.Desmocollins(DSCs)are members of the cadherin supergene family,which play an important role in cell-to-cell adhesion.Desmocollin-2(DSC2)is a member of the DSC family and has been found to be abnormally expressed in several types of cancers.It can affect tumor cell invasion and metastasis and participate in tumor progression.The differential expression and regulation of DSC2 in different human diseases provide potential targets for its treatment.To study and clarify the mechanism of DSC2 in tumor,to explore the specific targets of clinical examination and treatment can provide practical ideas for early treatment of disease.This article reviews the molecular structure of desmosome and DSC2,the relationship between DSC2 and tumor and its related molecular mechanism.
