摘要

自噬在乳腺癌(Breast cancer,BC)中的作用复杂,根据肿瘤的不同阶段,自噬发挥促进或抑制作用,这可能与不同的分子通路有关.miRNAs通过上调或下调细胞自噬,促进或抑制BC的进展,miRNAs还可以作为BC患者诊断和预后判断的生物标志物以及BC的潜在治疗靶点.通过调节Beclin-1蛋白影响自噬的发生进而抑制或促进BC的进展.H19 lncRNA通过不同分子通路在BC进展、增殖、转移和治疗中发挥关键作用.上调雌激素受体β(Estrogen receptor β,ERβ)能够促进BC细胞的自噬,抑制其迁移和侵袭.环状RNA circ-Dnmt1通过p53和AUF1通路抑制BC细胞增殖和存活,而KISS1通过调节自噬影响BC的远处转移.本文综述了自噬在乳腺癌中发挥作用的分子机制,为乳腺癌的防治提供参考.

Abstract

The role of autophagy in breast cancer is complex.It may play the role of promoting or inhibiting through different molecular pathways depends on the stage of the breast cancer(BC)A few miRNAs promote or inhibit the progression of BC by up-regulating or down-regulating autophagy,and can also be used as a biomarker for the diagnosis and prognosis of patients with BC,as well as the potential therapeutic targets for BC.Beclin-1 protein inhibits or promotes the progression of BC by affecting the occurrence of autophagy.H19 lncRNA plays a key role in the progression,proliferation,metastasis,and treatment of BC through different molecular pathways.Up-regulating ERβ can promote autophagy of BC cells and inhibit their migration and invasion.Circular RNA circ-Dnmt1 inhibits the proliferation and survival of BC cells through the p53 and AUF1 pathways.KISS1 affects distant metastasis of BC through autophagy.In this paper,the molecular mechanisms of autophagy in breast cancer was reviewed.It will be helpful for the prevention and treatment of breast cancer.
