中药活性成分诱导的铁死亡在骨肉瘤中的研究进展

摘要
骨肉瘤(Osteosarcoma,OS)是一种最常见的骨癌类型,具有高致残率和致死率的特点.因其发病机制复杂,OS的有效治疗一直是该领域内的难点.中药因其具有多重活性成分,在治疗OS方面展现出了独特优势.已有研究证实青蒿素类(双氢青蒿素、青蒿素)、类黄酮(补骨脂二氢黄酮)、姜黄素类(姜黄素、EF24)等中药活性成分能够通过铁死亡途径对骨肉瘤起治疗作用.铁死亡作为铁依赖的磷脂过氧化作用驱动的一种独特的细胞死亡方式,受到了多种细胞代谢途径的调控,与OS的发生、发展密切相关.本文归纳了铁死亡相关的理论和它在OS中的最新研究进展,围绕中药中不同活性成分在OS中调控铁死亡的分子机制进行了总结和讨论,以期为OS的预防和治疗提供参考.
Abstract
Osteosarcoma(OS)is the most common type of bone cancer,characterized by high disability and mortality rates.Due to its complex pathogenesis,effective treatment of OS has always been a challenge in this field.Traditional Chinese medicine has shown unique advantages in treating OS because of its multiple active ingredients.Previous studies have confirmed that active ingredients in traditional Chinese medicine,such as artemisinins(dihydroartemisinin,artemisinin),flavonoids(bavachin),and curcumins(curcumin,EF24),can exert therapeutic effects on osteosarcoma through the ferroptosis pathway.As a unique modality of cell death driven by iron-dependent phospholipid peroxidation,ferroptosis is regulated by multiple cellular metabolic pathways.They are closely related to the occurrence and development of OS.This article summarizes the theory related to ferroptosis and its latest research progress in OS,and summarizes and discusses the molecular mechanism of different active ingredients in traditional Chinese medicine in regulating ferroptosis in OS,in order to provide reference for the prevention and treatment of OS.
