基于Box-Behnken响应面设计优选乌蕨总黄酮的超声提取工艺

摘要
目的:利用Box-Behnken响应面设计法优选乌蕨总黄酮的超声提取工艺.方法:以乙醇浓度、液料比、超声提取温度作为考察因素,乌蕨总黄酮的提取量作为评价指标,采用Box-Behnken响应面设计法,优化乌蕨总黄酮提取工艺.结果:超声提取乌蕨总黄酮的最佳工艺条件:乙醇浓度34%,液料比30∶1(mL·g-1),超声提取温度72℃,超声时间40 min,在此条件下响应面预测值为46.132 mg·g-1,实测值为46.052 mg·g-1,RSD=1.61%.结论:基于Box-Behnken响应面设计方法实用可靠,效果较好,可以应用于乌蕨总黄酮的提取中,为乌蕨总黄酮的提取提供了一定的理论参考价值.
Abstract
Objective:To optimize the ultrasonic-assisted extraction process of total flavonoids from Stenoloma chusanum(L.)Ching using Box-Behnken response surface methodology.Methods:Single-factor experiments were conducted to explore three influencing factors:ethanol concentration,liquid-solid ratio,ultrasonic temperature.Regarding the extraction amount of total flavonoids as the evaluation index,the combination design of Box-Behnken was combined to optimize the optimal process of total flavonoids extraction.Results:The optimum process conditions for ultrasonic extraction of total flavonoids from Stenoloma chusanum were as follows:ethanol concentration 34%,liquid-solid ratio 30∶1(Ml·g-1),ultrasonic extraction temperature 72 ℃,ultrasonic time 40 min.Under these conditions,the predicted value of response surface was 46.132 mg·g-1,the measured average value was 46.052 mg·g-1,RSD=1.61%.Conclusion:The verification results show that this method is practical and reliable,and the effect is good.It can be applied to the extraction of total flavonoids from Stenoloma chusanum,which provides theoretical reference for the extraction of total flavonoids from Stenoloma chusanum.
