摘要

慢性肾脏病是近年来发现的一种发病率和死亡率逐渐上升的慢性疾病,各种病因导致的慢性肾脏病共同特征是肾纤维化.有研究表明上皮间质转化是肾纤维化的关键激活因素之一.上皮间质转化最初发现于胚胎正常发育过程中,随后在癌症的侵袭和迁移中以及器官纤维化过程中也发现了它的身影.目前的研究结果表明,上皮间质转化由多种信号通路调控,如TGF-β/Smad信号通路、Wnt/β-catenin信号通路等.但是上皮间质转化在肾纤维化的进展中,具体是如何通过上述信号通路发挥作用的还有待研究.此外,有研究表明上皮间质转化也可通过microRNA等小分子物质调控.本文主要从上皮间质转化的关键调控机制及其在肾纤维化中的作用来探讨通过靶向抑制上皮间质转化以防治肾纤维化的前景.

Abstract

Chronic kidney disease(CKD)is a chronic disease with increasing morbidity and mortality in recent years.The common feature of CKD caused by various causes is renal fibrosis.Studies have shown that epithelial mesenchymal transformation is one of the key activators of renal fibrosis.Epithelial mesenchymal transformation was first identified during normal embryonic development and also found in cancer invasion,migration and in organ fibrosis.Current findings suggest that epithelial mesenchymal transformation is regulated by a variety of signaling pathways,such as TGF-β/Smad signaling pathway and Wnt/β-catenin signaling pathway.In addition,studies have shown that epithelial mesenchymal transformation can also be regulated by microRNA and other small molecules.However,how epithelial mesenchymal transformation plays a role in the progression of renal fibrosis through the above signaling pathways remains to be studied.In this paper,the key regulatory mechanisms of epithelial mesenchymal transformation and its role in renal fibrosis were discussed to explore the prospect of preventing and treating renal fibrosis by targeting inhibition of epithelial mesenchymal transformation.
