摘要
目的:发展一种由CuI介导的简易高效的Reformatsky催化体系.方法:在便宜易得CuI的介导下,以乙腈/水混合溶剂为反应溶剂,以锌粉为催化剂催化碘代乙酸乙酯2a与亚胺1的Reformatsky反应.结果:在上述反应条件下,可高产率(95%)获得Reformatsky加成产物β-氨基酯3.产物结构经 1H NMR、13C NMR和HRMS确证.依据前期研究及相关文献,提出了可能的反应机理.结论:CuI介导的碘代乙酸乙酯2a与亚胺1的Reformatsky反应可放大至克级规模,有利于工业化生产.
Abstract
Objective:To develop a simple and efficient cuprous iodide-mediated catalytic system of Reformatsky reaction.Methods:The Reformatsky reaction of ethyl iodoacetate 2a with imines 1 was efficiently catalyzed by zinc powder and mediated by the cheap and easily available cuprous iodide with acetonitrile/water mixed solvent as the reaction solvent.Results:The Reformatsky addition products β-amino esters 3 can be obtained with high yield(up to 95%)under mild conditions and the structures of products 3 were characterized by 1H NMR、13C NMR and HRMS.In addition,according to the previous research and the related literature,the possible reaction mechanism was proposed.Conclusion:Mediated by cuprous iodide,the Reformatsky reaction of ethyl iodoacetate 2a with imines 1 can be scaled up to the gram scale,which is conducive to industrial production.
