组蛋白乳酸化修饰的研究进展

摘要
乳酸是细胞糖酵解的主要代谢产物,在能量供给和信号传导方面发挥重要作用,因此乳酸可作为反应底物参与多种疾病的发展.2019年首次报道组蛋白可发生乳酸化修饰,近年来组蛋白乳酸化修饰作为疾病发生的潜在机制被陆续报道.研究表明组蛋白乳酸化可改变肿瘤微环境促进肿瘤发生、参与脓毒血症发展、通过NF-κB信号通路促进脑衰老和阿尔茨海默病以及参与其他疾病发展.虽然大量研究显示组蛋白乳酸化会加快疾病发展进程,但也有研究报道组蛋白乳酸化同样发挥促进疾病恢复的作用.本文简要介绍组蛋白乳酸化修饰在临床疾病发展中的作用及其潜在机制,为组蛋白翻译后修饰研究提供理论基础.
Abstract
Lactic acid is the main metabolite of cellular glycolysis.It plays an important role in energy supply and signal transduction.As a reaction substrate,lactic acid can participate in the development of a variety of diseases.In 2019,it was first reported that histone lactylation could occur,and in recent years,histone lactylation was successively reported as a potential mechanism of disease occurrence.Studies have shown that histone lactate can change the tumor microenvironment to promote tumorigenesis,participate in the development of sepsis,promote brain aging and Alzheimer's disease through the NF-κB signaling pathway,and participate in the development of other diseases.Although a large number of studies have shown that histone lactate can accelerate the development of the disease,it has also been reported that histone lactate plays a role in promoting the recovery of the disease.This article briefly introduces the role and potential mechanism of histone lactate modification in the development of clinical diseases,and provides a theoretical basis for the study of histone post-translational modification.
