Analysis of the expression and clinical significance of POLE2 in lung adenocarcinoma based on biological information
Objective:To analyzed the difference in POLE2 expression between lung adenocarcinoma and adjacent tissues,as well as how POLE2 relates to clinic pathological characteristics and prognosis in lung adenocarcinoma patients,and pre-liminarily explored the involvement of POLE2 in the signaling pathway of lung adenocarcinoma,which are based on the pub-lic informatics information database.Methods:POLE2 expression and related clinical data of lung adenocarcinoma patients were download from TCGA database.The expression data of POLE2 in lung adenocarcinoma tissues and adjacent tissues were extracted by R4.0 software,and the expression difference between the two groups were compared.The expression of POLE2 was divided into high expression group and low expression group according to the median value,and the relationship between POLE2 expression and clinical pathological characteristics of lung adenocarcinoma were analyzed.Univariate and multivariate factor cox regression were used to analyze the relationship between POLE2 and the prognosis of lung adenocar-cinoma.Kaplan Meier method was used to draw a survival curve to compare the survival difference between the high and low expression groups of POLE2.The online GEPIA and OncoLnc databases were used for verification,and the signaling pathway involved in POLE2 in lung adenocarcinoma were analyzed by gene enrichment analysis(GSEA).Results:The expression of POLE2 in lung adenocarcinoma group was significantly higher than that in the adjacent group(P<0.05).The expression of POLE2 in lung adenocarcinoma patients was related to T stage(P<0.05)and age(P<0.05).POLE2(HR=1.212,95%CI:1.019-1.440,P<0.05)was an independent prognostic factor of lung adenocarcinoma.The KM survival curve of TCGA data-base showed that the survival time of POLE2 high expression group was lower than that of low expression group(P<0.05),which was consistent with GEPIA and OncoLnc results.POLE2 was mainly involved in EF2 protein family and G2M check-point signaling pathway in lung adenocarcinoma,and is related to cell cycle regulation.Conclusions:POLE2 is highly ex-pressed in patients with Lung adenocarcinoma,and the level of POLE2 is related to T stage and age.POLE2 expression can be used as one of the indicators to predict the prognosis of lung adenocarcinoma,and POLE2 may affect the progression of lung adenocarcinoma by affecting the conduction of various signaling pathways.
