Pig umbilical cord blood exosomes regulate myoblast proliferation and differentiation by delivering ssc-miR-451-5p targeting CDKN2D
[Objective]To study the functional regulation of umbilical cord blood exosomes(UCB-Exos)and ssc-miR-451-5p on C2C12 myoblasts,and to explore the regulatory effects of UCB-Exos and ssc-miR-451-5p on muscle development in pigs with intrauterine growth retardation(IUGR).[Method]Us-ing normal piglets and IUGR piglets as experimental materials,the regulatory effect of UCB-Exos on the proliferation and differentiation of C2C12 myoblasts was isolated and explored,the miRNA expression pro-file of UCB-Exos was constructed,the miRNA with the highest expression was screened,and the regula-tory effect of UCB-Exos and its highest expressed miRNAs on C2C12 myoblasts was explored by CCK8,EDU and qRT-PCR.The target gene with the highest miRNA expression was determined by diluciferase gene reporter experiment,and the target gene function was investigated by siRNA.[Result]UCB-Exos was able to significantly promote the proliferation and differentiation of C2C12 myoblasts in vitro(P<0.05),and there was a significant difference in the expression of miRNA in normal piglets and IUGR piglets UCB-Exos,and ssc-miR-451-5p was identified as the highest expressed miRNA in UCB-Exos,and ssc-miR-451-5p was expressed in normal piglets UCB-Exos than IUGR piglets.It promotes myoblast pro-liferation and inhibits differentiation,and directly targets CDKN2D(cell cycle regulatory gene)to carry out its functions.[Conclusion]IUGR is different from the expression of normal piglet UCB-Exos miRNAs,UCB-Exos can promote the proliferation and differentiation of C2C12 myoblasts,and the highest expres-sion of ssc-miR-451-5p in UCB-Exos can promote the proliferation and inhibit the differentiation of C2C12 myoblasts by targeting CDKN2D.
