Studies on the ultrasound-assisted enzymatic extraction and anti-inflammatory activity of abrus cantoniensis hance polysac-charide
Objecitve:On the basis of single factor experiment,response surface experiment was performed to optimize the extraction process of polysaccharide from Abrus cantoniensis Hance by ultrasound-assisted enzyme method.Its anti-inflammatory activity was analyzed by anti-inflammatory test in vitro.Methods:The effects of enzyme dosage,liquid-solid ratio,pH of enzymolysis,enzymolysis-ultrasonic power,enzymolysis-ultrasonic time,enzymolysis-ultrasonic temperature on the extraction rate of polysaccharide were investigated by single factor test.The extraction process of polysaccharide was optimized by response surface test.The contents of TNF-α,IL-1β,IL-6 and IL-10 were detected by ELISA method.Results:The optimized extraction conditions of Abrus cantoniensis Hance polysaccharide were as follows:cellulase content 2.8%,liquid-solid ratio 21∶1(mL/g),pH 5.0,enzymolysis-ultrasonic power 250 W,enzymolysis-ultrasonic temperature 49℃,enzymolysis-ultrasonic time 90 min.Under these conditions,the extraction rate of polysaccharide was 13.26%.Compared with the LPS group,the polysaccharide could decrease the contents of pro-inflammatory cytokines TNF-α,IL-1β and IL-6,and increase the contents of anti-inflammatory cytokine IL-10.Conclusion:The extraction efficiency of Abrus cantoniensis Hance polysaccharide by ultrasound-assisted enzyme method was high,and the extracted polysaccharide possessed strong anti-inflammatory activity.
