Effects of acupuncture on GLUT4 mRNA expression in skeletal muscle and glycolipid metabolism of prediabetic model rats
Objective To observe the effect of acupuncture on glucose transporter 4(GLUT4)mRNA expres-sion in skeletal muscle and glycolipid metabolism of prediabetes(PD)model rats,and explore the mechanism of acupuncture in improving insulin resistance in PD.Methods 40 SPF male Wistar rats were divided into blank group,model group,western medicine group,and acupuncture group by random number table method,with 10 rats in each group.The PD model of rats was established by feeding high sugar and high fat diet combined with intrape-ritoneal injection of 1%streptozotocin solution.After the model was successfully established,the acupuncture group was given acupuncture at Zhongwan(RN12)and Zusanli(ST36)(both sides)acupoints.After 4 weeks of interven-tion,the general conditions of rats in each group were observed,and the levels of fasting plasma glucose(FPG),2 hours postprandial blood glucose(2hPG),total cholesterol(TC),triglyceride(TG),low density lipoprotein choles-terol(LDL-C),fasting insulin(Fins)and the relative expression of GLUT4 mRNA in skeletal muscle tissue were de-tected.The insulin resistance index(IRI)and insulin sensitivity index(IAI)were calculated.The pathological chan-ges of pancreatic tissue were observed under microscope by HE staining.Results Compared with the blank group,the content of TG,TC,LDL-C,and 2hPG,Fins,IRI in the model group rats were significantly increased,while I AI and the relative expression of GLUT4 mRNA in skeletal muscle tissue were significantly reduced,and the differences were statistically significant(P<0.05 or P<0.01).Compared with the model group,the content of TG,TC,LDL-C,and 2hPG,Fins,IRI in the acupuncture group rats were significantly reduced,while I AI and the relative expression of GLUT4 mRNA in skeletal muscle tissue were significantly increased,and the differences were statistically signifi-cant(P<0.05 or P<0.01).The observation results under microscope by HE staining showed that acupuncture can reduce the degree of pancreatic tissue damage in PD model rats and make the arrangement of pancreatic islet cells tend to be neat.Conclusion Acupuncture can effectively improve insulin resistance in PD model rats,and its mechanism may be related to reducing blood glucose and blood lipid levels,increasing the expression of GLUT4 mRNA in skeletal muscle tissue,thereby accelerating the speed of glucose transport and alleviating the disorder of glycolipid metabolism.
