目的 建立膜果麻黄的鉴别方法.方法 采用基源鉴别、性状鉴别、显微鉴别及理化鉴别方法对药用植物膜果麻黄进行鉴别,获取药材形态组织学的主要鉴别特征;以麻黄碱及伪麻黄碱作为膜果麻黄的指标成分,采用薄层色谱(TLC)法及超高效液相色谱(UPLC)法进行定性鉴别,分析色谱图特征.结果 膜果麻黄茎圆柱形,表面常纵裂成椭圆形网眼,茎顶端极度扭曲;膜质鳞叶3裂稀2裂,先端锐尖,反曲;髓呈三角形;鳞叶表皮细胞垂周壁波状弯曲;气孔内陷,保卫细胞顶面观呈电话听筒形;花粉粒呈椭圆形,花瓣细胞排列紧密,纤维壁厚,其上附有细小的颗粒状晶体;茎横切面皮层较宽,可见细小草酸钙晶体,形成层及木质部呈三角形.TLC及UPLC结果显示,膜果麻黄未发现麻黄碱及伪麻黄碱成分.结论 本研究建立的鉴别方法及试验结果高效、准确、可靠,可为膜果麻黄的真伪鉴别提供参考.
Objective To establish a method for the identification of Ephedra przewalskii Stapf.Methods The origin identification,character identification,microscopic identification and physical and chemical identification methods were used to identify the medicinal plant Ephedra przewalskii Stapf to obtain the main identification features of morphology and histology of the medicinal material.Ephedrine and pseudoephedrine were used as the index com-ponents of Ephedra przewalskii Stapf.Thin-layer chromatography(TLC)and ultra performance liquid chromatogra-phy(UPLC)were used for qualitative identification and chromatographic characteristics were analyzed.Results The stem of Ephedra przewalskii Stapf is cylindrical,often longitudinally split into elliptical mesh on the surface,and the top of the stem is extremely twisted.Membranous scale leaves are 3-lobed,seldom 2-lobed,apex sharp,re-curved;the pith is triangular;the vertical wall of epidermal cells of squamous leaf is wavy.The stomata were invagi-nated,and the top surface of the guard cells was shaped like a telephone handset.The pollen grains are oval,the petal cells are closely arranged,the fiber wall is thick,and there are fine granular crystals on it.The transverse sec-tion of the stem is wide,small calcium oxalate crystals can be seen,cambium and xylem are triangular.TLC and UPLC results showed that ephedrine and pseudoephedrine were not found in Ephedra przewalskii Stapf.Conclusion The identification method and experimental results established in this study are efficient,accurate and reliable,and can provide reference for the identification of Ephedra przewalskii Stapf.
