化学计量学分析党参中金属元素及其健康风险评估

目的 建立电感耦合等离子体质谱(ICP-MS)测定党参中20种金属元素含量的方法,并对党参潜在的健康风险进行评估.方法 党参药材采用微波消解进行前处理,利用ICP-MS测定党参中20种金属元素的含量,采用聚类分析、主成分分析(PCA)和偏最小二乘判别分析(PLS-DA)等方法对不同产地党参样品中的元素含量进行差异性分析,通过计算每日最大可耐受摄入量(EDI)、靶标危害系数(THQ)、危险指数(HI)进行风险评估.结果 所测党参样品中,2批重金属砷超标,2批重金属镉超标;通过化学计量学分析发现,20种元素中,铝、钒、铁、钴、铅、钾、铊、钙、铜、锰、硒、锶、镁等13种元素是区别不同产地党参样品的差异标志物.同时,健康风险评估模型EDI、THQ、HI结果显示,36批党参样品对人体健康不存在致癌风险.结论 党参对人体健康不存在致癌风险,但重金属超标现象仍然存在,应加强党参的种植、加工环节,以提高党参用药安全.
Stoichiometric analysis of metallic elements in Radix Codonopsis Pilosulae and its health risk assessment
Objective To establish an inductively coupled plasma mass spectrometry(ICP-MS)method for the determination of 20 metal elements in Radix Codonopsis Pilosulae,and to evaluate its potential health risks.Me-thods Radix Codonopsis Pilosulae was pre-processed by microwave digestion.The contents of 20 metal elements in Radix Codonopsis Pilosulae were determined by ICP-MS.The differences of elements in the samples from different origin were analyzed by cluster analysis,principal component analysis(PC A)and partial least squares discriminant analysis(PLS-DA).Risk assessment was performed by calculating the maximum endurable daily intake(EDI),tar-get hazard quotients(THQ),and hazard index(HI).Results Among the samples of tested,the heavy metal arsenic in 2 batches and the heavy metal cadmium in 2 batches exceeded the standard.Through stoichiometric analysis,it was found that among the 20 elements,13 elements such as aluminum,vanadium,iron,cobalt,lead,potassium,thal-lium,calcium,copper,manganese,selenium,strontium and magnesium were the differential markers for distinguis-hing the samples from different areas.At the same time,the results of health risk assessment models EDI,THQ and HI showed that 36 batches of Codonax ginseng samples had no carcinogenic risk to human health.Conclusion Ra-dix Codonopsis Pilosulae has no carcinogenic risk to human health,but the problem of excessive heavy metals still exists.The planting and production of codonopsis codonopsis should be strengthened to improve the safety of drug use.
