Differences between two methods in detection of SARS-CoV-2 and diarrhea viruses in sewage
Objective To evaluate the differences between two enrichment methods in detection of nucleic acids from severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and various diarrheal viruses and access the epidemiological trends of the viruses in sewage water,so as to provide theoretical support for the formulation of prevention and control plans.Methods The polyethylene glycol precipitation method and anions membrane method ultrafiltration method were used to concentrate and detect SARS-CoV-2,norovirus(Nov),rotavirus(RV),sapovirus(SaV),adenovirus(AdV)and human astrovirus(HAstV)in sewage and the results were analyzed.Results The anions membrane method ultrafiltration method was better than the polyethylene glycol precipitation method for concentration of AdV.The polyethylene glycol precipitation method had higher concentration efficiency for Nov,RV and SaV.There is no statistically significant difference between the two methods for enriching of HAstV.The detection rate of the SARS-CoV-2 varied over time.After March 2,the concentration effect for Orf1ab and N genes by polyethylene glycol precipitation method was better than that of anion membrane method.Conclusions The two methods showed different concentration effects for different viruses in sewage and should be selected according to the needs of the work.
