Genetic analysis on VP1 gene of coxsackievirus B5 isolated from HFMD cases in Yunnan province,2022
Objective To perform molecular typing of 7 strains of coxsackievirus B5(CVB5)isolated from hand,foot and mouth disease(HFMD)cases in Yunnan province in 2022 and analyze the genetic characteristics of the VP1 gene.Methods The stool samples from HFMD cases through HFMD laboratory surveillance system of Yunnan province in 2022 were used for virus isolation.For positive samples,reverse transcription polymerase chain reaction(RT-PCR)and sequencing were used for VP4/VP2 region first,and then for VP1 gene.The phylogentic tree of VP1 gene was constructed using MEGA 7.0 software for the genetic analysis.Results Seven CVB5 strains were detected from the stool samples of HFMD patients in Yunnan province,2022.The similarity analysis based on VP1 gene sequences showed that the 7 CVB5 strains were most closed to the Henan strain COXB5-Henan-2010.The nucleotide(nt)and amino acid(aa)similarities among the 7 strains were 98.00%-99.65%and 99.30%-100%,respectively.The nt and aa similarities between the 7 strains and Henan strain were 87.36%-88.68%and 82.96%-84.85%,respectively.The phylogentic analysis showed that CVB5 could be divided into 4 genotypes(genotype A-D).The Yunnan strains belonged to genotype C and clustered into a sole lineage.The nt diversities among 4 genotypes was 18.90%-22.30%which were more than 75%in similarity and met the requirement of international enteroviruses(EVs)genotype assignment.Conclusions Seven CVB5 strains were identified by HFMD surveillance system in Yunnan province in 2022.Although the number was relatively lower,CVB5 was proved to be one of the causative pathogens of HFMD.The virus did exist in Yunnan province and clustered into a sole lineage.Etiological surveillance and molecular analysis for EVs other than EV-A71 and CVA16,especially CVB5 should be enhanced.
