Objective To establish a long-term gastritis model of Mongolian gerbils infected with Helicobacter pylori(Hp),and to evaluate the effect of Ligilactobacillus salivarius LI01 on the model.Methods A total of 48 SPF healthy Mongolian gerbils were initially divided into three groups of normal control group(NC group),Hp model group(Hp group),and Ligilactobacillus salivarius LI01 intervention group(Hp+LI01 group)with 16 Mongolian gerbils in each group.From week 1 to 5,the NC group was continuously gavaged with saline,while the Hp and Hp+LI0l groups were gavaged with Hp to induce gastritis.At week 30,6 Mongolian gerbils from each group were randomly euthanized to confirm successful Hp colonization and the establishment of gastritis model.Subsequently,the NC and Hp groups were gavaged with saline for 4 weeks,while the Hp+LI01 group was gavaged with Ligilactobacillus salivarius L101 for 4 weeks.During this period,growth indicators such as fur condition and body weight were recorded.At week 39 and 40,comet assays and hematoxylin-eosin(HE)staining were performed on histopathological sections to analyze the effects of Hp and Ligilactobacillus salivarius LI01 on the Mongolian gerbils.Results After 5 weeks of Hp administration,gerbils in the Hp and Hp+LI01 groups showed fur abnormalities,including dullness,piloerection,and hair loss.However,Ligilactobacillus salivarius LI01 intervention improved the fur condition in the Hp+LI01 group,which became comparable to the NC group.At week 34,body weights of different groups showed significantly different(F=325.52,P<0.001),with the Hp+LI01 group weighing(83.39±3.37)g,significantly higher than that of the Hp group(P<0.001).Comet assay results showed significant differences in comet length,tail length,tail moment,Olive tail moment,and DNA percentage in comet tail among the NC,Hp and Hp+LI01 groups(F=95.59,80.89,33.97,31.74 and 46.34,all P<0.001).All the above indicators in Hp group were significantly greater than those of NC group and Hp+LI01 group(all P<0.001).At week 40,the results of gastric histological examination showed that 4 gerbils were in stageⅠ,3 in stage Ⅱ,and 3 in stage Ⅲ of gastritis in the Hp group.In the Hp+LI01 group,2 gerbils were in stage 0,5 in stage Ⅰ,and 3 in stage Ⅱ.Conclusions Gastric lavage with Hp successfully induces a gastritis model in Mongolian gerbils,while Ligilactobacillus salivarius LI01 demonstrates a potential for improvement and repair of the associated gastric damage.
