Study on a simple and improved perfusion technique of isolated rat kidney
Objective To explore a simple and improved technique of isolated renal perfusion in rats,through the improvement of renal perfusion equipment.Methods Twelve SD male rats in No-vember 2021 were selected as the study subjects and randomly divided into conventional perfusion group and improved perfusion group,with 6 rats in each group.The abdominal aorta was used as the perfusion inflow channel,and the renal vein and ureter of rats were used as the outflow channels of per-fusate.The complications of perfusion were treated according to the situation during the experiment.The perfusion effect of different perfusion methods was evaluated comprehensively by calculating the successful rate of perfusion in the renal artery,the amount of leakage fluid during perfusion and the a-mount of blood and urine specimens collected from the rats by different perfusion methods.Results The peritoneal fluid leakage in the improved perfusion group was lower than that in the conventional perfusion group,and the difference was statistically significant(P<0.05).Urine volume in improved perfusion group was higher than that in conventional perfusion group,and the difference was statistical-ly significant(P<0.05).The venous fluid volume in the improved perfusion group was lower than that in the conventional perfusion group,and the difference was not statistically significant(P>0.05).Conclusions The modified perfusion method allows for more efficient flow of perfusate through the kidney and less leakage of perfusate into the peritoneal cavity,as well as easier collection of blood and urine specimens,thus providing a simpler and more effective experimental procedure.
