Expression of BMP-2 and BMP-7 in non-muscular invasive bladder cancer patients and their clinical prognostic significance
Objective To investigate the expression and prognostic significance of recombi-nant human bone morphogenetic protein-2(BMP-2)and recombinant human bone morphogenetic pro-tein-7(BMP-7)in patients with non-muscular invasive bladder cancer.Methods A total of 118 pa-tients with non-muscular invasive bladder cancer who underwent transurethral resection in our hospital from January 2016 and January 2018 were selected as the research subjects.The expression levels of BMP-2 and BMP-7 protein in bladder cancer tissue(bladder cancer group)and paracancer tissue(pa-racancer group)were determined by immunohistochemical method.The relationship between BMP-2 and BMP-7 protein expression levels and clinicopathological characteristics of patients with non-muscu-lar invasive bladder cancer were investigated by x2 test.Multivariate Cox regression analysis was used to explore the prognostic risk factors of patients with non-muscular invasive bladder cancer.Results The positive expression rate of BMP-2 and BMP-7 protein in the bladder cancer group was lower than that in the adjacent group(all P<0.05).The protein expression of BMP-2 and BMP-7 was not signif-icantly correlated with age and gender(all P>0.05).It was significantly correlated with tumor maxi-mum diameter,TNM stage,invasion depth,pathological grade,lymph node metastasis and recurrence(allP<0.05).Moreover,the maximum tumor diameter≥5 cm,the higher the TNM stage,the dee-per the invasion depth,the higher the pathological grade,the lower the positive expression rates of BMP-2 and BMP-7 in patients with lymph node metastasis and recurrence(all P<0.05).The 5-year survival rate of BMP-2 and BMP-7 positive patients with bladder cancer was significantly higher than that of BMP-2 and BMP-7 negative patients(75.0%vs.41.0%,77.8%vs.41.5%,all P<0.05).The 5-year survival rate of bladder cancer patients with maximum tumor diameter ≥ 5cm,TNM stage Ⅲ-Ⅳ,lymphatic vascular invasion,high pathological grade,recurrence,BMP-2 negative and BMP-7 negative were significantly shortened(all P<0.05).Cox regression analysis showed that TNM stage Ⅲ to Ⅳ,high pathological grade,negative BMP-2 and negative BMP-7 were independent risk factors for the prognosis of patients with bladder cancer(all P<0.05).Conclusions The ex-pression of BMP-2 and BMP-7 protein in cancer tissues of patients with non-muscle invasive bladder cancer are decreased,and the two are negatively correlated with the progression of non-muscle invasive bladder cancer.Negative expression of BMP-2 and BMP-7 are risk factors for poor prognosis of non-muscle invasive bladder cancer.
