Identification of bacterial flora in the first urine of patients with primary urethritis and male patients with non-idiopathic urethritis
Objective To analyze the bacterial flora in the first urine of patients with primary urethritis and non-idiopathic urethritis by using the cloning library method.Methods Sixty-four first urine specimens of male patients with urethritis(primary urethritis and non-idiopathic urethritis)were collected in our hospital from January 2019 to June 2020,based on the 16S ribosomal RNA gene clone library method analyzes the bacterial flora.In addition,conventional PCR methods are used to detect neisseria gonorrhoeae,chlamydia trachomatis,mycoplasma genitalium,mycoplasma hominis,ure-aplasma urealyticum or ureaplasma parvum.Results Among the 64 urine specimens,42 were suc-cessfully analyzed by the clonal library method,from the specimens,2 781 clones were evaluated and 95 bacterial phylogenetic types were detected.Cloning library method:Neisseria gonorrhoeae was de-tected from 6 specimens,and it was the main bacterial species in 5 specimens.Mycobacterium genita-lia was detected from 5 specimens,and it was the advantage of 3 specimens strains.Ureaplasma urea-lyticum was detected in 2 specimens.Haemophilus influenzae and Neisseria meningitidis were detected in 8 and 1 specimens.Gardnerella vaginalis was detected in 10 specimens.TMA method in conven-tional PCR detected chlamydia trachomatis from 15 specimens.Neisseria gonorrhoeae was detected in 8 specimens,and it was the dominant strain in 5 specimens,and two samples accounted for 100%of the flora.In conventional PCR,the TaqMan method detected Mycobacterium genitalium in 12 samples and accounted for more than 50%of the flora in 2 samples.Neisseria gonorrhoeae was detected in 1 sample and accounted for 100%.Conclusions The cloning library method can detect the occupancy rate of each species in the flora,and may become a new method for the analysis of male urethritis bacteria.
