Study on the relationship between the expression of FLOT1 and PSMB4 in bladder cancer and the clinicopathologic features and prognosis
Objective To explore the relationship between the expression of Flotillin 1(FLOT1)and proteasome β subunit type 4(PSMB4)in bladder cancer(BC)and the clinicopatho-logical characteristics and prognosis of BC patients.Methods One-hundred BC patients who were treated in our hospital from June 2017 to June 2022 were selected as the study subjects.The expression of FLOT1 and PSMB4 proteins in BC tissue was detected by fluorescent quantitative PCR and immuno-histochemistry,the relationship between the expression of FLOT1,PSMB4 and the clinicopathological features of BC was analyzed.Kaplan-Meier curve was used to analyze the relationship between FLOT1,PSMB4 and the prognosis of BC patients.Cox risk proportional regression model was used to evaluate the prognostic factors of patients with BC.Results The expression of FLOT1 and PSMB4 in BC cancer tissue was significantly higher than that in adjacent tissues(t=32.095,24.318,all P<0.05).The positive rates of FLOT1 and PSMB4 in cancer tissues were significantly higher than those in adjacent tissues(x2=16.822,17.321,all P<0.001).The expression of FLOT1 and PSMB4 in cancer tissue is correlated with tumor length,pathological grade,lymph node metastasis,and TNM staging(all P<0.05).The 5-year survival rate of the positive FLOT1 expression group was lower than that of the neg-ative FLOT1 expression group(P<0.05).The 5-year survival rate of PSMB4-positive expression group was lower than that of negative expression group(P<0.05).The results of multi-factor analysis showed that TNM stage,FLOT1 and PSMB4 positive expression were independent risk factors for the prognosis of BC patients(all P<0.05).The results of receiver operating characteristic(ROC)curve showed that the area under the curve(AUC)of combined prediction was significantly higher than that of single prediction(all P<0.05).Conclusions The expression of FLOT1 and PSMB4 proteins in BC tissue is increased,which is related to the pathological grade,lymph node metastasis,TNM stage and prognosis of BC,and is an independent risk factor for the prognosis of BC patients.The combination of FLOT1 and PSMB4 protein has a certain predictive value for the occurrence of poor prognosis of BC.
