Relationship between expression of serum C-type lectin receptor and its downstream effector CARD9 and clinicopathological features and prognosis of rectal cancer patients
Objective An attempt is made in this paper to investigate the relationship between the expression of serum C-type lectin receptor(CLR)and its downstream effector caspase recruitment domain protein 9(CARD9)and the clinicopathological features and prognosis of rectal cancer patients.Methods A total of 98 patients with rectal cancer at the Sinopharm Tongmei General Hospital from March 2018 to March 2020 were selected as the subjects(assigned to the observation group),and 45 healthy volunteers who underwent physical examinations at the hospital during the same period were selected and assigned to the control group.The expression of serum soluble dendritic cell-specific intercellular adhesion molecular 3-grabbing nonintegrin(sDC-SIGN)and CARD9 was detected by using the ELISA method,and the relationship between the expression of serum sDC-SIGN and CARD9 and clinicopathological features of rectal cancer patients was analyzed.Pearson correlation analysis was used to explore the relationship between serum sDC-SIGN and CARD9.All enrolled rectal cancer patients were followed up for a period of 3 years after surgery,and the 3-year overall survival(OS)rate was calculated.The Kaplan-Meier method was used to draw survival curves and analyze the relationship between serum sDC-SIGN and CARD9 expression and prognosis of rectal cancer patients.The prognostic factors of rectal cancer patients were explored using multivariate Cox regression analysis.The predictive value of serum sDC-SIGN and CARD9 in postoperative survival of rectal cancer patients was analyzed using ROC curve analysis.Results Compared with the control group,the expression level of serum sDC-SIGN is decreased and the expression level of CARD9 is increased in the observation group,with statistically significant differences(P<0.05).The Pearson correlation analysis shows that there is a significant negative correlation between serum sDC-SIGN and CARD9 expression in rectal cancer patients(r=-0.743 and P<0.05).Compared with rectal cancer patients without lymph node metastasis,the expression level of serum sDC-SIGN in patients with lymph node metastasis is lower,while the expression level of CARD9 is higher,with statistically significant differences(P<0.05).The 3-year OS rate of the sDC-SIGN low expression group is lower than that of the sDC-SIGN high expression group,and the 3-year OS rate of the CARD9 high expression group is lower than that of the CARD9 low expression group,with statistically significant differences(P<0.05).The results of multivariate Cox regression analysis show that the differentiation degree,TNM stage,lymph node metastasis,serum sDC-SIGN,and CARD9 are independent risk factors affecting the prognosis of rectal cancer patients(P<0.05).The area under the ROC curve(AUC)of serum sDC-SIGN combined with CARD9 for predicting 3-year postoperative survival in rectal cancer patients is 0.896,which is higher than the efficacy of single detection(P<0.05).Conclusion The expression levels of serum sDC-SIGN and CARD9 are closely related to the occurrence of rectal cancer,and the low expression of serum sDC-SIGN and high expression of serum CARD9 suggest poor postoperative prognosis in rectal cancer patients,which may become a new serological marker for the diagnosis and prognosis prediction of rectal cancer.
