Efficacy of Ruangan Zhuyu formula on nonalcoholic steatohepatitis in high-fat diet-induced mice
Objective To observe the efficacy and anti-inflammatory activity of Ruangan Zhuyu formula in the treatment of high-fat diet-induced nonalcoholic steatohepatitis(NASH)in mice,and to provide data support for its clinical application.Methods Forty-two C57BL/6 mice were randomly divided into model group,normal group,Silibinin Capsules group,Ruangan Zhuyu formula low,medium,and high dosage groups,seven mice in each group.The NASH model was induced in mice by feeding with high-fat diet for 14 weeks.After successful modelling,Ruangan Zhuyu formula each dose groups and the Silibinin Capsules group were given 1 ml drug solution every day by intragastric administration for 13 weeks on the basis of high-fat diet.The changes of body mass in each group were recorded.The mice were executed 2 weeks after the administration was stopped,and blood samples were collected to detect the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),laminin(LN),hyaluronic acid(HA),interleukin(IL)-2,IL-4,IL-6,IL-10 and interferon a(IFN-a).Hematoxylin-eosin(HE),oil-red,and Masson staining were used to observe the hepatic steatosis,fat droplet accumulation,and hepatic fibrosis.Results Compared with the model group,the body mass of mice in Ruangan Zhuyu formula group decreased significantly(P<0.05).The serum AST level in Ruangan Zhuyu formula high-dose group decreased significantly(P<0.05),the serum ALT level in Ruangan Zhuyu formula medium and high-dose groups decreased significantly(P<0.05),the level of IL-4,IL-6,IL-10 and IFN-a decreased significantly(P<0.05).The level of HA in Ruangan Zhuyu formula group decreased significantly(P<0.05).The results of HE,oil red and Masson staining showed that liver fibrosis and liver inflammation were significantly reduced in mice in Ruangan Zhuyu formula medium and high-dose groups.Conclusion Ruangan Zhuyu formula can effectively reduce the body mass of mice induced by high-fat diet,and improve the liver function and liver fibrosis of mice.
