UPLC法测定小柴胡颗粒中柴胡皂苷b1、b2

Determination of saikosaponin b1 and saikosaponin b2 in Xiaochaihu Granules by UPLC

目的 建立UPLC法测定小柴胡颗粒中柴胡皂苷b1、b2.方法 使用Acquity UPLC BHE C18 色谱柱(100 mm×2.1 mm,1.7 μm);流动相:水-乙腈,梯度洗脱;检测波长:254 nm;体积流量:0.3 mL/min;柱温:30℃;进样量3 μL.结果 柴胡皂苷b1、b2 线性范围分别为 0.006~0.374 mg、0.003~0.214 mg,平均回收率分别为 98.97%、99.35%,RSD值分别为 1.82%、1.16%.结论 所建立方法准确、稳定,重现性好,可用于小柴胡颗粒的质量控制和评价.
Objective To develop an UPLC method for determination the saikosaponin b1 and saikosaponin b2 in Xiaochaihu Granules.Methods The analysis was applied on Acquity UPLC BHE C18 column(100 mm×2.1 mm,1.7 μm).The mobile phase consisted of water and acetonitrile with gradient elution.The detection wavelength was set at 254 nm,flow rate was 0.3 mL/min,column temperature was 30℃,and volume of injection was 3 μL.Results Saikosaponin b1 and saikosaponin b2 showed good linear relationships of 0.006—0.374 mg and 0.003—0.214 mg,whose average recoveries were 98.97%and 99.35%with RSD values of 1.82%and 1.16%.Conclusion The established method is accurate,stable,and has good reproducibility,and can be used for quality control and evaluation for Xiaochaihu Granules.
