Influence of characteristic massage on inflammation and the expression of SOCS1 and TLR4 proteins in a spinal cord nerve ligation rat model
Objective This study aimed to explore the anti-inflammatory and analgesic mechanisms of characteristic massage in neuropathic pain(NP).Methods Thirty SPF-grade SD rats were randomly allocated to the normal group,model group,massage group,sham massage group,and sham surgery group,with 6 rats in each group.Except that the normal group was not treated and the sham surgery group was exposed to L5 nerve,the remaining three groups were subjected to spinal nerve ligation(SNL)to establish the NP rat model.Starting from the first day after modeling.The massage group received characteristic massage treatment,while the sham massage group,gentle touch.The interventions lasted for 7 days.On the day before modeling and days 1,3,and 7 after modeling,the paw withdrawal latency(PWL)to thermal stimuli and the paw withdrawal threshold(PWT)to mechanical stimuli were measured in all groups.After 7 days of intervention,the ex-pression levels of suppressor of cytokine signaling 1(SOCS1),Toll-like receptor 4(TLR4),and nuclear factor-κB(NF-κB)in the spinal dorsal horn were detected by Western blot,and the level of interleukin-6(IL-6)in the spinal dorsal horn tissue was measured by ELISA.Results On the day before modeling,no significant difference was observed in PWT and PWL a-mong the groups(P>0.05).On days 1,3,and 7 after modeling,compared with the normal and sham surgery groups,the PWT and PWL of the sham massage group,massage group,and model group were all significantly decreased(P<0.05).Additionally,the PWT and PWL of the massage group were significantly increased on days 3 and 7 after modeling as com-pared with the model and sham massage groups(P<0.05).Compared with the normal and sham surgery groups,the lev-els of SOCS1 protein in the sham massage group,massage group,and model group were significantly increased(P<0.05).Compared with the model and sham massage groups,the levels of SOCS1 in the massage group were significantly increased(P<0.05).Compared with the normal and sham surgery groups,the levels of TLR4,NF-κB,and IL-6 in the spinal dorsal horn of the sham massage group,massage group,and model group were significantly increased(P<0.05).Compared with the model group,the levels of TLR4,NF-κB,and IL-6 in the massage group were significantly decreased(P<0.05).Con-clusion The characteristic massage can improve NP induced by SNL,and its mechanism may be related to the upregulation of SOCS1 protein expression in the spinal dorsal horn,the downregulation of TLR4 and NF-κB protein expression through negative feedback,and the reduction of pro-inflammatory cytokine IL-6 production.
