Expression levels of serum miR-182-3p and miR-342-3p and their correlation with insulin resistance in pregnant women with gestational diabetes mellitus
Objective To investigate the expression levels of serum miR-182-3p and miR-342-3p in patients with gestational diabetes mellitus(GDM)and their correlation with insulin resistance(IR).aiming to provide a reference for the clinical di-agnosis and treatment of GDM.Methods A total of 85 GDM patients who were hospitalized and delivered at Xingtai Central Hospital from October 2021 to October 2023 were selected as the study group,and 80 healthy pregnant women who came to the hospital for prenatal check-ups and delivery during the same period were selected as the control group.A quantitative re-al-time fluorescent quantitative polymerase chain reaction(qRT-PCR)was used to detect the levels of miR-182-3p and miR-342-3p in the serum of all study participants.A fully automatic biochemical analyzer was adopted to detect the levels of fast-ing blood glucose(FBG)and fasting insulin(FINS)in the study participants,and the homeostasis model assessment of in-sulin resistance(HOMA-IR)was calculated.Pearson correlation analysis was used to analyze the correlation between serum miR-182-3p,miR-342-3p levels,and HOMA-IR.Multiple linear regression analysis was employed to analyze the related in-fluencing factors of HOMA-IR in GDM patients.Furthermore,the receiver operating characteristic(ROC)curve was used to analyze the diagnostic value of serum miR-182-3p and miR-342-3p levels for GDM.Results Compared with the control group,the levels of FBG,FINS,HOMA-IR,glycated hemoglobin(HbA1c),and relative expression levels of miR-182-3p and miR-342-3p in study group were higher than those in control group,with statistical significance(P<0.05).Pearson a-nalysis showed that the levels of serum miR-182-3p and miR-342-3p in GDM patients were positively correlated with HO-MA-IR levels(r=0.392,0.485,all P<0.001).Multiple linear regression analysis indicated miR-182-3p and miR-342-3p are influencing factors of HOMA-IR in GDM patients(P<0.05).The area under curve(AUC)for diagnosing GDM using serum miR-182-3p,miR-342-3p,or both combined were 0.841,0.833,and 0.916,respectively,with the combination be-ing significantly superior to either miR-182-3p or miR-342-3p alone(Z=3.629,2.740,all P<0.05).Conclusion The lev-els of serum miR-182-3p and miR-342-3p in GDM patients are elevated and have a certain correlation with IR,which can provide new biomarkers for the early diagnosis and treatment of GDM.
