Relationship between YBX1 and FGFR2 expression in nasopharyngeal carcinoma tissues and patient pathological features and prognosis
Objective To explore the relationship between the expression of Y-box binding protein-1(YBX1)and fibroblast growth factor receptor 2(FGFR2)in nasopharyngeal carcinoma tissues and patient pathological features and prognosis,ai-ming to provide a reference for clinical treatment. Methods A total of 136 patients with nasopharyngeal carcinoma who were initially diagnosed and treated at the Affiliated Hospital of Shaanxi University of Chinese Medicine from February 2018 to February 2021 were selected as the study participants.Nasopharyngeal carcinoma tissue specimens before treatment were included in the nasopharyngeal carcinoma group(n=136),and adjacent non-tumor tissue specimens were included in the control group(n=136).Spearman correlation analysis was used to assess the correlation between YBX1 and FGFR2,and multivariate Cox regression analysis was adopted to identify factors affecting the prognosis of patients with nasopharyngeal carcinoma. Results Compared with the control group, the positive expression rates of YBX1 and FGFR2 were significantly higher in the nasopharyngeal carcinoma group(P<0.05).Spearman correlation analysis indicated a positive correlation be-tween YBX1 and FGFR2 expression in nasopharyngeal carcinoma tissues(rs=0.462,P<0.05).Patients with N stage 2-3,TNM stage Ⅳ,lymph node metastasis, and low tumor differentiation had significantly higher positive expression rates of YBX1 than those with N stage 0-1,TNM stage Ⅱ-Ⅲ,absence of lymph node metastasis, and medium/high tumor dif-ferentiation(P<0.05).Similarly,patients with N stage 2-3,TNM stage Ⅳ,lymph node metastasis, and low tumor dif-ferentiation had significantly higher positive expression rates of FGFR2 than those with N stage 0-1,TNM stage Ⅱ-Ⅲ,absence of lymph node metastasis, and medium/high tumor differentiation(P<0.05).Among the 136 patients with naso-pharyngeal carcinoma,110 patients survived after a 3-year follow-up post-discharge,with a total survival rate of 80.88%(110/136).Multivariate Cox regression analysis results demonstrated that N stage 2-3(HR=3.080,95%CI:1.409-6.733),TNM stage Ⅳ(HR=2.309,95%CI:1.324-4.029),positive YBX1(HR=3.300,95%CI:1.826-5.965),and positive FGFR2(HR=4.540,95%CI:1.997-10.322)were independent influencing factors for the prognosis of pa-tients with nasopharyngeal carcinoma(P<0.05).Conclusion The positive expression rates of YBX1 and FGFR2 in naso-pharyngeal carcinoma tissues are significantly increased and are associated with the clinical pathological features of patients with nasopharyngeal carcinoma.Patients with positive YBX1 and FGFR2 expression have a poorer prognosis, which can serve as supplementary prognostic markers for nasopharyngeal carcinoma.
