Research on Optimization of Fermentation Medium for Lactobacillus plantarum AR307
Lactic acid bacteria have received widespread attention and research due to their various physiological functions such as enhancing immunity,promoting digestion,and reducing serum cholesterol.To further enhance the proliferation density of fermented lactic acid bacteria,this experiment used Lactobacillus plantarum AR307 as the experimental object,MRS medium as the basic medium,and optimized the medium using single factor and response surface methodology.The paper studies the effects of carbon sources,nitrogen sources,and inorganic salts on bacterial growth,and determines the optimal ratio using OD600 as the evaluation index.The results show that the optimal carbon source is maltose,with an added amount of 3.5%;The optimal nitrogen source is yeast extract powder,casein peptone,and beef extract powder,with a ratio of 1∶1∶1 and an added amount of 2.21%;The optimal inorganic salt is potassium dihydrogen phosphate,with a dosage of 0.75%.Under these optimized conditions,after 24 h of fermentation,the OD600 value of Lactobacillus plantarum AR307 reached 6.4,and the bacterial density was improved,providing a theoretical basis for the industrial fermentation of AR307.
