摘要

文章旨在探究不同浓度玫瑰精油对D-gal诱导的皮肤成纤维细胞衰老的影响.设立空白对照组、D-gal组、D-gal+玫瑰精油组.用β-gal染色检测阳性细胞数目、EdU-594 染色检测细胞增殖能力和划痕实验检测细胞迁移能力.β-gal染色结果显示,D-gal+玫瑰精油组与对照组(D-gal组)比较,衰老染色阳性细胞数减少(p<0.001);EdU-594 染色结果显示,与对照组相比,D-gal+玫瑰精油组增殖细胞数增多(p<0.01);细胞迁移实验结果显示,D-gal+玫瑰精油组细胞迁移率增加(p<0.001).可见,玫瑰精油可以减少D-gal模型中细胞衰老数目,提高皮肤成纤维细胞的增殖能力,增加皮肤成纤维细胞的迁移能力.

Abstract

The aim of this study was to investigate the effects of rose essential oil at different concentrations on D-gal induced skin fibroblast senescence.There were blank control group,D-gal group and D-gal+ rose essential oil group.β-gal staining was used to detect the number of positive cells,EdU-594 staining was used to detect the proliferation of cells and scratch assay was used to detect the migration of cells.The β-gal staining results showed that the number of senescence staining positive cells decreased in the D-gal+ rose essential oil group compared with the control group(D-gal group)(p<0.001);EdU-594 staining showed that compared with the control group,the number of proliferating cells in D-gal+ rose essential oil group increased(p<0.01);The results of cell migration experiment showed that compared with the control group,the cell migration rate of D-gal+ rose essential oil group increased(p<0.001).Therefore,rose essential oil can reduce the number of cell senescence in D-gal model,improve the proliferation ability of skin fibroblasts,and increase the migration ability of skin fibroblasts.
