The Effects of Tanshinone ⅡA on β1/Smads Signaling Pathway in Hepatic Stellate Cells of Rats Induced by Angiotensin Ⅱ
Objective:To investigate the effects of Tanshinone ⅡAon β1/Smads signaling pathway in hepatic stellate cells of rats induced by Angiotensin Ⅱ and discuss the mechanism of Tanshenone ⅡA in treating hepatic fibrosis.Methods:Cultured HSC,and then treated them with Angiotensin Ⅱ(1 × 10-5mol/L) or Angiotensin Ⅱ(1 × 10-5mol/L) and Tanshinone ⅡA (25μg /ml).Treated cells and control groups were incubated respectively for 48 hours.Relative mRNA expression of TGF-β1,Smad2 and Smad4 were assayed by quantitative real-time polymerase chain reaction.Relative proteins expression of TGF-β1 and Smad4 were investigated by Western blot.Results:Angiotensin Ⅱ significantly up-regulated both the mRNA and proteins expression of TGF-β1,Smad2 and Smad4 in HSC,while Tanshinone ⅡA significantly attenuated both the mRNA and proteins expression of TGF-β1,Smad2 and Smad4 in HSC treated by Angiotensin Ⅱ.Conclusions:This study demonstrated that Tanshinone ⅡA can significantly down-regulate the relative mRNA/protein expression of TGF-β1,Smad2 and Smad4.It is suggested that Tanshinone ⅡA may be a therapeutic option for the prevention of hepatic fibrosis.
