丹参酮ⅡA对血管紧张素Ⅱ诱导的大鼠肝星状细胞TGF-β1/Smads通路的影响

目的:研究丹参酮ⅡA对体外培养的血管紧张素Ⅱ诱导的肝星状细胞(hepatic stellate cell,HSC)转化生长因子β1 (TGF-β1)/Smads通路中TGF-β1、Smad2、Smad4的mRNA和蛋白表达的作用,探讨丹参酮ⅡA治疗肝纤维化的机理.方法:对大鼠肝星状细胞分别使用血管紧张素Ⅱ(1×10-5mol/L)及血管紧张素Ⅱ(1×10-5mol/L)联合丹参酮ⅡA(25μg/ml)干预48h,分别用real-time PCR及western blot方法检测各组细胞TGF-β1、Smad2、Smad4的mRNA及蛋白表达.结果:血管紧张素Ⅱ上调肝星状细胞TGF-β1、Smad2、Smad4的mRNA及蛋白表达(P<0.01),而丹参酮ⅡA对血管紧张素Ⅱ在肝星状细胞中的这种诱导作用具有明显的抑制(P<0.01).结论:丹参酮ⅡA抗肝纤维化的机制之一可能是通过对肝星状细胞TGF-β1/Smads通路的调控实现的.
The Effects of Tanshinone ⅡA on β1/Smads Signaling Pathway in Hepatic Stellate Cells of Rats Induced by Angiotensin Ⅱ
Objective:To investigate the effects of Tanshinone ⅡAon β1/Smads signaling pathway in hepatic stellate cells of rats induced by Angiotensin Ⅱ and discuss the mechanism of Tanshenone ⅡA in treating hepatic fibrosis.Methods:Cultured HSC,and then treated them with Angiotensin Ⅱ(1 × 10-5mol/L) or Angiotensin Ⅱ(1 × 10-5mol/L) and Tanshinone ⅡA (25μg /ml).Treated cells and control groups were incubated respectively for 48 hours.Relative mRNA expression of TGF-β1,Smad2 and Smad4 were assayed by quantitative real-time polymerase chain reaction.Relative proteins expression of TGF-β1 and Smad4 were investigated by Western blot.Results:Angiotensin Ⅱ significantly up-regulated both the mRNA and proteins expression of TGF-β1,Smad2 and Smad4 in HSC,while Tanshinone ⅡA significantly attenuated both the mRNA and proteins expression of TGF-β1,Smad2 and Smad4 in HSC treated by Angiotensin Ⅱ.Conclusions:This study demonstrated that Tanshinone ⅡA can significantly down-regulate the relative mRNA/protein expression of TGF-β1,Smad2 and Smad4.It is suggested that Tanshinone ⅡA may be a therapeutic option for the prevention of hepatic fibrosis.
