Screening the Experimental Animal Model of Renal Interstitial Fibrosis from Multi Angle
Objective:To compare the models established by adenine irrigation and by unilateral ureteral ligation (UUO) so as to obtain a better experimental animal model of renal interstitial whose biochemical,immunohistochemical and pathological indicators meet the standard better.Methods:Experimental rats were divided into normal group,Adenine group and UUO group.The Adenine group and UUO group were given adenine irrigation and unilateral ureteral ligation (UUO) to establish the experimental animal model of renal interstitial.Observe the urine protein quantitation,Serum creatinine and urea nitrogen,HE,Masson staining,pathological changes,E-cadherin、β-catenin and α-SMA of every group.Results:Compared with the normal group,the 24-h urine protein quantitation,Serum creatinine and urea nitrogen increased in Adenine group(P <0.01),and in UUO group,the urine protein quantitation had a significant increase(P <0.05),while Serum creatinine and urea nitrogen increased as well(P <0.01).Compared with Adenine group,the 24-h urine protein quantitation of UUO group creased remarkably(P <0.05).Compared with the normal group,the average optical density of E-cadherin decreased significantly (P < 0.05)and Alpha-SMA beta and-catenin average optical density values increased significantly (P < 0.01,P < 0.05).Compared with Adenine group,β-catenin in UUO group decreased greatly (P < 0.05).Pathologic revealed that renal tubular injury in Adenine group was more serious than that in UUO group.Conclusion:Animal Model of Renal Interstitial Fibrosis established in UUO group is better than Adenine group in biochemistry,immunohistochemistry and pathology.
