雪上一枝蒿药材及其制剂发挥镇痛作用的治疗窗研究

目的:考察雪上一枝蒿药材及其制剂发挥镇痛作用的治疗窗范围,并比较它们之间的毒性大小.方法:采用经典的急性毒性试验方法进行雪上一枝蒿及其制剂的急性致死量试验,醋酸扭体法进行镇痛药效试验,采用SPSS17.0统计软件计算急性毒性的LD50、LD5和发挥镇痛药效的ED50、ED95,求得各组别发挥镇痛作用的治疗指数(TI),安全系数(SF),以此评价雪上一枝蒿药材及其制剂发挥镇痛作用的有效性和安全性.结果:各组别发挥镇痛作用的治疗窗有所不同,药材醇提液、总生物碱溶液、总生物碱微乳发挥镇痛作用的治疗指数TI分别为5.250、6.825、10.460,安全系数SF分别为1.178、1.650、1.788.结论:雪上一枝蒿总生物碱微乳与总生物碱溶液、雪上一枝蒿药材醇提液相比,其治疗窗较宽.三者的治疗窗大小为:总生物碱微乳＞总生物碱溶液＞药材醇提液.
The Analgesic Effect Study on Therapeutic Window of Aconitum Brachypodum Diels and its Preparation
Objective:To study the range of therapeutic window on the analgesic effect of Aconitum brachypodum Diels and its preparation,and compare their toxicity.Method:To take research on the acute dosis lethalis of Aconitum brachypodum Diels and its preparation with the method of classical acute toxicity test,and on the analgesic effect with the classical acetic acid writhing method.Obtained the therapeutic index (TI) of LD50,LD5,ED50,ED95 and the safety factor(SF) of the analgesic effect of Aconitum brachypodum Diels and its preparation by using SPSS17.0 statistical software in order to evaluate its efficacy and safety.Result:Each therapeutic window is different.TI of the alcohol extract from Aconitum brachypodum Diels,of the solutions and of the microemulsion of total alkali was 5.250,6.825 and 10.460 respectively,while SF of them was 1.178,1.650 and 1.788 respectively.Conclusion:The therapeutic window of the analgesic effect on Aconitum brachypodum Diels and its preparation is:microemulsion of the total alkali ＞ solutions of the total alkali ＞ alcohol extract of Aconitum brachypodum Diels.
