Determining the Contents of Five Hydrosoluble Ingredients in Kangerxin Capsule
Objective:To establish a new quantitative method for determining five hydrosoluble Iingredients (Sodium danshensu;protocatechuic acid;protocatechuic aldehyde;rosmarinic acid;salvianolic acid B) in Kangerxin capsule by HPLC.Method:Chromatographic separation was carried out on Thermo syncronis-C18 column (4.6 mm ×250 mm,5μm) under gradient elution with methanol(A phase)-1% glacial-Acetic acid (B phase) as mobile phase at a flow rate of 1.0ml · min-1.The injection volume was 10 μL and the detection was adopted UV detector at 281 nm.Result:These five components were separated basically within 55 minutes with good linearity.The regression equations were Y =642.88X + 1.98,r =0.9999 (for Sodium danshensu),Y=4372213.83X + 805.16,r =0.9999 (for protocatechuic acid),Y =1245713.26X + 4412.89,r =0.9999 (for protocatechuic aldehyde),Y =1713766.30X + 1387.01,r =0.9999 (for rosmarinic acid),Y =1209485.54X +29336.62,r =0.9998(for salvianolic acid B).The average recoveries were:Sodium danshensu 99.8% (RSD 1%);protocatechuic acid 98.5% (RSD 0.9%);protocatechuic aldehyde 98.3% (RSD 1.6%);rosmarinic acid 103.8% (RSD 0.7%);salvianolic acid B 98.3% (RSD 0.8%).Conclusion:The HPLC method has accuracy,reliability,and repeatability.It is suitable to be used to control the quality of for Kangerxin capsule.
