心衰宁颗粒对慢性心力衰竭大鼠心肌组织转化生长因子β1的影响

目的:探讨心衰宁颗粒对慢性心力衰竭大鼠心肌转化生长因子β1(TGFβ1)表达的影响.方法:将SD大鼠随机分为正常组、模型组、卡托普利组、心衰宁大、中、小剂量组,除正常组外,其余组采用阿霉素腹腔注射制作慢性心力衰竭大鼠模型,正常组注射等量生理盐水,每周1次,造模6周结束后,分别给予药物干预,正常组及模型组给予生理盐水,干预3周.干预结束后,超声心动图检测心功能,大鼠血清学检测proBNP、TGFβl水平;qRT-PCR、Western blot检测检测TGFβ1基因及蛋白表达水平.结果:与正常组比较,模型组左心室舒张末期内径(LVEDD)和左心室收缩末期内径(LVESD)明显增加(P<0.01),左室射血分数(LVEF)降低(P<0.O1),血清TGF-β1、proBNP明显升高(P<0.01),TGF-β1基因及蛋白表达较正常组明显升高(P<0.01);与模型组比较,心衰宁颗粒中、大剂量组能降低LVEDD、LVESD(P <0.05),提高慢性心衰大鼠LVEF(P <0.01),降低血清TGF-β1、proBNP水平(P<0.01),降低TGF-β1基因及蛋白表达水平(P<0.05).结论:心衰宁颗粒具有改善心功能,干预心肌重塑、抗心肌纤维化作用,其机制可能与抑制TGF-β1水平有关.
Effect of Xinshuaining Granule on Transforming Growth Factor Beta 1 of Rats with Chronic Heart Failure
Objective:To discuss the influence of expression of Xinshuaining Granule on transforming growth factor beta 1 (TGF-β1) of rats with chronic heart failure.Methods:SD rats were divided into a normal group,a model group,a group with Captopril,a group with high-dose Xinshuaining granule,a group with medium-dose Xinshuaining granule and a group with low-dose Xinshuaining granule.Except the normal group,SD rats in other groups were induced to get chronic heart failure by Intraperitoneal injection with adriamycin,while the rats in the normal group were injected with equivalent normal saline once a week for 6 weeks.After successfully establishing the model,each group was given corresponding drug intervention respectively for 3 weeks.Then,echocardiography was used to test the cardiac function.The levels of proBNP and TGF-β1 in rats serum were tested.Using the qRT-PCR and western blot detected the gene and protein expression level of TGF-β1.Methods:Compared with the normal group,the left ventricular end diastolic dimension (LVEDD) and left ventricular end systolic dimension(LVESD) increased significantly (P <0.01).The left ventricular ejection fraction (LVEF) reduced (P < 0.01),the serum TGF-β1 and proBNP increased significantly (P < 0.01) and the gene and protein expression level of TGF-β1 increased significantly (P < 0.01).Compared with the model group,the group with medium-dose Xinshuaining granule and the group with high-dose Xinshuaining granule could reduce LVEDD and LVESD (P < 0.05),improved the LVEF of rats with chronic heart failure (P <0.01),reduced the level of serum TGF-β1 and proBNP(P <0.01) and reduced the gene and protein expression level of TGF-β1 (P < 0.05).Conclusion:Xinshuaining granule can improve heart function,interpose cardiac remodeling and resist myocardial fibrosis.Its mechanism may be concerned with the inhibiting the TGF -β1 level.
