Analysis of microbiological characteristics of the buffer zone at the inlet of Hongfeng Lake based on high-throughput sequencing
In this study,Hongfeng Lake was selected as the research object.It is an important drink-ing water source in Guiyang,and soil samples of 4 main branches flowing into the buffer zone of the lake mouth under different ecological environments were collected.HiSeq sequencing was used to se-quence the 16S rRNA V4 region of soil bacteria and the ITS1 region of fungi,combined with soil en-vironmental factors.The composition characteristics and diversity of microbial community were ana-lyzed in order to provide reference and basis for restoring vegetation ecology in the buffer zone of Hongfeng Lake.The results showed that there were 53 phyla and 813 genera of bacteria in the four buffer zones.There are 6 phyla and 406 genera in the species,and the composition of each buffer zone is similar at the phyla level.The common dominant bacterial phyla are Proteobacteria,Acidobacte-ria,Cyanobacteria,etc.The dominant fungal phyla are Ascomycota,Basidiomycota and Chytridiomycota.At the bacterial genus level,the content of nitrifying and denitrifying bacteria(7.13%)in the soil of Maiweng River buffer zone was higher than that of other three buffer zones.The diversity index of soil bacteria in the four buffer zones was HL>MX>MW>YC.The diversity index of fungi was YC>MX>HL>MW.The results of correlation analysis showed that bacterial diversity index was significantly pos-itively correlated with total nitrogen(n=12,r=0.567,p<0.05),and fungal community diversity index was significantly positively correlated with total salt content(n=12,r=0.622,p<0.05).Therefore,dif-ferent plant composition has an impact on the composition and diversity of soil microorganisms in the buff-er zone of lake shore,and human reclamation activities also play an important role.
