采用抗坏血酸还原氯金酸制备纳米金,在钴离子的存在下,半胱氨酸诱导纳米金团聚,溶液颜色由红色变为蓝色,据此建立一种半胱氨酸测定新方法.讨论了纳米金-钴离子比色探针的制备条件,探究了测定半胱氨酸的实验条件.结果表明:纳米金-钴离子比色探针测定半胱氨酸的最佳反应时间为 20 min、缓冲溶液pH为 3.0,该方法测定半胱氨酸的线性回归方程为 A725/A520 =0.3965 C+0.0133(C 单位为 μM),R2=0.9882,线性范围为1×10-7~1×10-6 mol/L,方法检出限为 2.2×10-8 mol/L.采用标准加入法对健康人的尿样进行回收率测定,回收率为92.8%~95.8%.文中创建了一种简便测定半胱氨酸的方法.
Preparation of Nanometer Gold-cobalt Ion Colorimrtric Probe and Determination of Cysteine
In this paper,ascorbic acid was used to reduce chlorauric acid to prepare gold nanoparticles.In the presence of cobalt ion,cysteine induced gold nanoparticles to agglomerate and the color of the solution changed from red to blue.A new method for determination of cysteine was established.The preparation conditions of gold-cobalt ion colorimetric probe were discussed and the experimental conditions for determination of cysteine were investigated.The results show that:The optimal reaction time and pH of buffer solution were 20 min and 3.0 respectively.The linear regression equation for the determination of cysteine was A725/A520 =0.3965C+0.0133(C:μM),R2=0.9882.The linear range was 1×10-7 mol/L to 1×10-6 mol/L,and the detection limit was 2.2×10-8 mol/L.The recovery rate of urine samples from healthy people was determined by standard addition method,and the recovery rate was 92.8%~95.8%.A simple method for the determination of cysteine was developed.
