Determination of Protein Binding Rate of Chlorantraniliprole in Rat Plasma Using Equilibrium Dialysis Device Combined with UPLC-MS/MS
To determine the protein.binding rate of chlorantraniliprole in rat plasma.The equilibrium dialysis method was used to simulate the binding process of chlorantraniliprole with plasma proteins in rats,and the UPLC-MS/MS method was used to detect the concentration inside and outside the membrane of the equilibrium dialysis device.The results showed that there was a good linear relationship between the internal and external dialysis fluids in the range of 50~2500 ng/mL and 0.5~250 ng/mL.There was a good accuracy and precision within and between batches.The extraction recovery rate was from 94.4%to 104.2%.The accuracy of stability samples for internal and external dialysis fluids was from 85%to 115%.Within the low,middle and high concentration(150、800、2000 ng/mL)of chlorantraniliprole,the protein binding rate of rat plasma was 98.5%,97.9%and 97.6%respectively.The chlorantraniliprole had high protein binding rate in rat plasma.
