Synthesis and Crystal Structure of 15-Ketone of Prostaglandin
Using para-phenyl benzoyl colchicine as the raw material,(3aR,4R,5R,6S)-2-oxo-4-[(E)-3-oxooct-1-en-1-yl]cyclohexyl-2H-cyclopenta[b]furan-5-yl[1,1'-biphenyl]-4-carboxylate was synthesized through oxidation and Wittig-Horner reaction with a total yield of 72.22%.It's structure was confirmed by nuclear magnetic resonance spectroscopy,and mass spectrometry,and it's crystal structure was detrmined using X-ray single crystal diffraction.The results show that the tested block crystal a belongs to the orthorhombic crystal system,with a space group of P212121 and cell parameters:a=8.2476(2)Å,b=12.7324(3)Å,c=22.8279(7)Å;α=β=γ=90.00°;cell volume V=2397.20(11)Å3,asymmetric unit in the cell Z=4,R1=0.0384,ωR2=0.1039,S=1.028.This route have few impurities,simple post-treatment,and high yield.
