玄参不同"发汗"时间总黄酮含量变化的研究

目的 建立玄参总黄酮提取及含量测定方法,并对不同"发汗"时间的样品进行测定,探讨其变化规律。方法 以芦丁为对照品,采用紫外-可见分光光度法对玄参总黄酮含量进行测定;利用L9(34)正交试验优选总黄酮的提取工艺。结果 优选总黄酮提取工艺为:25 mL 60%的乙醇溶液于90℃回流提取40 min。不同"发汗"时间玄参总黄酮含量范围为1。20%~1。63%,平均含量为1。39%,其中,"发汗"0d的总黄酮含量最低,为1。20%;"发汗"11 d的含量最高,为1。63%。结论 优选的玄参总黄酮提取工艺及检测方法简单、稳定、合理、可行,可用作玄参总黄酮含量的测定;玄参总黄酮含量受"发汗"时间的影响,存在较大差异,随着"发汗"时间的延长,整体呈上升趋势。本研究结果为玄参药材"发汗"的进一步研究奠定了基础。
A Study on the Change of Total Flavonoids Content in Radix Scrophulariae at Different Sweating Time
Objective To establish a method for the determination of total flavonoid content of Radix Scro-phulariae,to measure the variation patterns of samples with different"sweating"time,and to discuss its chan-ging rule.Methods Using rutin as the reference substance,the content of total flavonoids was determined by UV visible spectrophotometry.Using orthogonal experimental design L9(34)to optimize the extraction process of total flavonoids.Results The optimal extraction process for total flavonoids was as follows:25mL of 60% etha-nol was refluxed at 90 ℃ for 40 minutes.The total flavonoid content ranged from 1.20% to 1.63% at different sweating time,with an average content of 1.39%.Among them,the content of sweating for zero days was the lowest,which was 1.20%,while the content of sweating for 11 days was the highest,which was 1.63%.Con-clusions The optimized extraction process for total flavonoids in Radix Scrophulariae is simple,stable,reason-able,and feasible,which can be used for the determination of total flavonoids in Radix Scrophulariae.The total flavonoids in Radix Scrophulariae is influenced by the sweating time.There are significant differences in their content.As the"sweating"time prolongs,the overall trend shows an upward trend.The research results lay the foundation for further research on the sweating of Radix Scrophulariae medicinal herb.
